目的:通过研究去卵巢骨质疏松症大鼠股骨中BMP2和Smad1的蛋白表达,探讨摘除卵巢致肾虚骨质疏松症的病理机制,并研究左归丸的作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制骨质疏松症模型,采用左归丸对实验大鼠治疗12周,以福善美片作为阳性对照组,正常组、假手术组作为标准对照组,模型组作为空白对照。用ELISA法检测骨质疏松症大鼠股骨BMP2和Smad1的蛋白表达。结果:正常大鼠股骨中存在BMP2和Smad1的蛋白表达。模型组大鼠股骨中BMP2和Smad1的蛋白表达水平有明显的下降。左归丸组、福善美组用药12周后,与模型组相比较,左归丸组和福善美组可显著上调股骨中BMP2和Smad1的蛋白表达水平。结论:1股骨中BMP2和Smad1的蛋白表达下降可能是导致骨质疏松症发生的机理之一;2左归丸能够上调BMP2和Smad1的蛋白表达,表明左归丸对骨质疏松症有一定的防治作用。
OBJECTIVE: To investigate the protein expression of BMP2 and Smad1 in the femur of ovariectomized rats with osteoporosis, and to explore the pathological mechanism of osteoporosis induced by kidney removal in ovariectomized rats and the mechanism of Zuogui Pill. Methods: Osteoporosis model was induced by ovariectomized female rats. The rats were treated with Zuogui Pill for 12 weeks. Foshenmei tablets were taken as positive control group, normal group and sham operation group as standard control group. Model group as a blank control. The protein expression of BMP2 and Smad1 in the femur of osteoporosis rats was detected by ELISA. Results: BMP2 and Smad1 protein expression existed in the normal femur. The protein expression of BMP2 and Smad1 in the femur of the model group decreased significantly. Compared with model group, Zuoguiwan group and Fosamax group could significantly up-regulate the expression of BMP2 and Smad1 in femur in Zuoguiwan group and Fosamax group for 12 weeks. Conclusion: 1 The decreased BMP2 and Smad1 protein expression in femur may be one of the mechanisms leading to osteoporosis.2 Zuogui pill can up-regulate BMP2 and Smad1 protein expression, indicating that Zuo Gui Wan has some osteoporosis Prevention and treatment.
