从浙江温州盘菜上获得芜菁花叶病毒温州分离物(TuMV-WZ),病毒提纯后,经电镜观察,可发现大量长约700nm的线形粒子。利用免疫捕捉RT-PCR,通过特异性引物对TuMV-WZ的HC-Pro基因进行PCR扩增,经电泳可见1.5kb的特异条带。PCR扩增产物经过纯化后进行序列测定,序列长度为1449个核苷酸,编码482个氨基酸。其核苷酸序列与已报道的TuMV的其他分离物(Canada,UKl和Japanese)同源率分别为83.9%、96.5%和94.0%,氨基酸同源率分别为97.3%、98.8%和99.0%。
The isolate of TuMV-WZ was obtained from Panax quinquefolium in Wenzhou, Zhejiang Province. After purification by electron microscopy, a large number of linear particles about 700 nm in length were found. Using immunocapture RT-PCR, specific primers were used to amplify the HC-Pro gene of TuMV-WZ. The specific band of 1.5kb was observed by electrophoresis. The PCR product was purified and sequenced. The length of the PCR product was 1449 nucleotides and encoded 482 amino acids. Their nucleotide sequences were 83.9%, 96.5% and 94.0%, respectively. The amino acid homologies were 97.3%, 98.8% and 99.0%, respectively, for the other isolates of TuMV (Canada, UKl and Japanese).
