本文研究的目的为探讨MC在大肠癌PDT增效作用中对癌细胞周期DNA含量的影响。方法为在癌细胞培养的基础上,用超低浓度的MC增效;PDT后采用流式细胞技术,对癌细胞的周期和DNA含量进行分析,数据经SPLM软件包处理。其结果是MC可使SW480细胞G1期细胞百分比显著增高;S期细胞、PI指数显著降低;PDT后,PSD+MC组各期细胞与对照组比细胞百分比无显著变化,与PSD组比,G1期细胞明显升高,G2+M期细胞、PI指数显著降低;MC可使DNA含量减少,PDT后DNA含量相对增加。其结论为超低浓度MC通过增加静止期细胞数量来达到增效作用。
The purpose of this study was to investigate the effect of MC on the DNA content of cancer cell cycle in PDT synergism of colorectal cancer. The method is based on the cultivation of cancer cells, using ultra-low concentration of MC to enhance efficiency; PDT after the use of flow cytometry, the cycle of cancer cells and DNA content analysis, data processed by SPLM software package. As a result, the percentage of G1 phase cells in SW480 cells was significantly increased by MC; the number of cells in S phase and the PI index were significantly decreased; after PDT, the percentage of cells in each phase of PSD+MC group was not significantly different from that of the control group. Compared with the PSD group, G1 phase cells After a significant increase, G2+M phase cells, PI index decreased significantly; MC can reduce the DNA content, DNA content after PDT relative increase. The conclusion is that the ultra-low concentration of MC achieves a synergistic effect by increasing the number of cells in the stationary phase.
