目的探讨RNA干扰乏氧诱导因子1α(HIF1α)对人肺腺癌细胞SPCA1摄取18F脱氧葡萄糖(FDG)的影响。方法构建干扰HIF-1α质粒pSUPERHIF-1α,采用荧光定量逆转录聚合酶链反应(RTPCR)、Western印迹法观察有氧与乏氧状态下RNA干扰后HIF1α的变化,用γ计数仪检测其对SPCA1细胞摄取18F-FDG的影响。结果RNA干扰HIF-1α,在乏氧和常氧状态下致mRNA表达分别下降76%和64%,空载体对照组分别为5%和0%;在乏氧状态下,蛋白质的表达下降,SPCA-1细胞对18F-FDG的摄取明显降低(P<0.05)。结论在乏氧状态下,RNA干扰HIF-1α明显降低SPCA-1细胞对18F-FDG的摄取。
Objective To investigate the effect of HIF1α RNA interference on uptake of 18G deoxyglucose (FDG) by human gastric adenocarcinoma cell line SPCA1. Methods Plasmid pSUPERHIF-1α, which interferes with HIF-1α, was constructed. Fluorescent quantitative reverse transcriptase-polymerase chain reaction (RTPCR) and Western blotting were used to observe the changes of HIF1α after RNA interference in aerobic and hypoxia conditions. Effect of cellular uptake of 18F-FDG. Results The mRNA expression of HIF-1α was decreased by 76% and 64% in hypoxia and normoxia, respectively, compared with 5% and 0% in empty vector control group, respectively. The expression of HIF-1α was decreased in hypoxia and SPCA -1 cells uptake of 18F-FDG was significantly lower (P <0.05). Conclusion In hypoxia, RNA interference of HIF-1α significantly decreased the uptake of 18F-FDG in SPCA-1 cells.
