目的观察野牡丹提取物对血栓形成、血小板聚集及血小板cAMP、cGMP和一氧化氮(NO)含量的影响。方法在颈外静脉注射给药5min后,用血栓生成仪观察野牡丹提取物对大鼠颈总动脉血栓形成的影响,测定5min平均堵塞率;全血与药物孵育5min,用全血血小板聚集仪观察全血血小板聚集情况;用ELISA法检测血小板cAMP和cGMP含量;用硝酸还原酶法观察血小板释放NO情况。结果野牡丹提取物5mg/kg促进大鼠颈总动脉血栓形成,堵塞率为(86.56±1.54)%。200、400、800mg/L野牡丹提取物均能诱导全血血小板聚集,并减少血小板cAMP、cGMP和NO生成。结论野牡丹提取物可促进大鼠血栓形成和血小板聚集,并能减少血小板cAMP、cGMP和NO含量。
Objective To observe the effect of wild peony extract on the thrombosis, platelet aggregation and platelet cAMP, cGMP and nitric oxide (NO) content. Methods The rats were sacrificed at 5 min after external jugular vein injection. The thrombus formation of carotid artery was observed by thrombus generator. The average clogging rate was measured at 5 min. The whole blood was incubated with drug for 5 min. The whole blood platelet aggregation The aggregation of platelets in whole blood was observed. The levels of cAMP and cGMP in platelets were detected by ELISA. The NO release in platelets was observed by nitrate reductase method. Results Wild Peony extract 5mg / kg to promote carotid artery thrombosis in rats, the blocking rate was (86.56 ± 1.54)%. 200,400,800 mg / L wild peony extract can induce whole blood platelet aggregation, and reduce platelet cAMP, cGMP and NO generation. Conclusion Wild Peony extract can promote thrombosis and platelet aggregation in rats, and can reduce platelet cAMP, cGMP and NO content.
