目的建立与分析2型糖尿病(T2DM)维吾尔医异常黒胆质证大鼠模型血浆代谢组学变化特点。方法建立T2DM维吾尔医异常黑胆质证大鼠模型并采集血浆样本,测试核磁共振氢谱,对数据进行多变量分析。结果单纯的异常黑胆质证组大鼠血浆中葡萄糖和乳酸含量下降的同时VLDL、LDL和肉碱的含量明显增加(P<0.05);病证结合的T2DM维吾尔医异常黑胆质证组大鼠血浆中多种氨基酸(丙氨酸、异亮氨酸、、缬氨酸、亮氨酸、酪氨酸、谷氨酸、苯丙氨酸、甲基组氨酸、甘氨酸)以及乳酸、丙酮酸、糖蛋白、柠檬酸、肌酸酐、β-羟丁酸、乙酰乙酸、乙酸、肌酸、肉碱、鲨肌醇、VLDL、LDL的含量均降低,而葡萄糖和牛磺酸含量明显增加(P<0.05)。结论蛋白质代谢、苯丙氨酸和酪氨酸代谢通路、糖酵解通路和脂代谢通路在病证结合的T2DM维吾尔医异常黑胆质证模型大鼠体内发生了明显的变化;血浆代谢组学变化在一定程度上反映病证结合的T2DM维吾尔医异常黑胆质证大鼠模型体内发生的病理变化过程。
Objective To establish and analyze the characteristics of plasma metabonomics changes in type 2 diabetes mellitus (T2DM) Uighur abnormalities. Methods T2DM Uygur anomalous savda syndrome rat model was established and plasma samples were collected. Nuclear magnetic resonance (1H) spectra were measured and multivariate analysis was performed on the data. Results Compared with the control group, the contents of VLDL, LDL and carnitine were significantly increased (P <0.05) in the plasma of rats with abnormal savda syndrome. (Alanine, isoleucine, valine, leucine, tyrosine, glutamic acid, phenylalanine, methyl histidine, glycine) as well as lactate, pyruvate, Glycoprotein, citric acid, creatinine, β-hydroxybutyric acid, acetoacetic acid, acetic acid, creatine, carnitine, scyllo-inositol, VLDL and LDL decreased, while the contents of glucose and taurine increased ). Conclusions Protein metabolism, phenylalanine and tyrosine metabolic pathways, glycolytic pathways and lipid metabolism pathways were significantly different in the T2DM Uighur salamander syndrome model rats.
