目的探讨竹根提取物抗Ⅰ型变态反应的药理作用及物质基础。方法通过对前期实验得到的一系列竹根提取物及3个单体化合物,应用Schultz-Dale反应实验,观察竹根提取物对豚鼠离体回肠的作用。结果竹根乙酸乙酯提取物有效部位(YY35、YY51-59)、氯仿提取物有效部位(C14、C23-30)以及单体成分(SUB-1)抑制Schultz-Dale反应结果分别为(0.23±0.20)g、(0.09±0.05)g、(0.09±0.08)g、(0.28±0.26)g和(0.49±0.26)g;C14对卵蛋白(OVA)的抑制率为(49.96%±0.12%)(3 min)和55.60%±0.11%(5 min),C23-30为(57.72%±0.11%)(3 min)和(55.77%±0.10%)(5 min);C14、C23-30对抑制组胺导致的张力增加结果为(94.39%±0.27%)、84.52%±0.31%。结论 YY35、YY51-59、C14、C23-30及SUB-1具有显著的抗Ⅰ型变态反应的作用。
Objective To investigate the pharmacological effects and material basis of bamboo root extract against type Ⅰ allergic reaction. Methods The Schultz-Dale reaction experiments were conducted on a series of bamboo root extracts and three monomer compounds obtained from previous experiments to observe the effect of bamboo root extract on the isolated ileum of guinea pigs. Results The Schultz-Dale inhibition results of effective fractions (YY35, YY51-59), C14, C23-30 and SUB-1 of ethyl acetate extract of bamboo root were (0.23 ± 0.20) g, (0.09 ± 0.05) g, (0.09 ± 0.08) g, (0.28 ± 0.26) g and (0.49 ± 0.26) g, respectively. The inhibition rate of C14 on OVA was (49.96% ± 0.12% (57.72% ± 0.11%) for 3 min and (55.77% ± 0.10%) for 5 min for C23-30 (5 min) and 55.60% ± 0.11% Histamine-induced increases in tension were (94.39% ± 0.27%) and 84.52% ± 0.31%. Conclusion YY35, YY51-59, C14, C23-30 and SUB-1 have significant anti-type I allergic response.
