目的探讨UPA、UPAR基因及蛋白表达与鼻咽癌的发生、发展及浸润转移之间的关系。方法应用原位杂交技术和流式细胞术分别检测鼻咽部慢性炎症患者20例、鼻咽癌无转移患者20例、鼻咽癌仅颈部淋巴结转移患者20例和鼻咽癌淋巴结及远处器官转移患者20例等4组患者组织中UPA、UPAR mRNA及蛋白的表达水平。结果各组中UPA、UPAR mRNA和UPA、UPAR蛋白质表达水平由高到低依次为:鼻咽癌远处器官转移组、鼻咽癌仅淋巴结转移组、鼻咽癌无转移组和慢性鼻咽炎症组。组间表达水平比较UPA mRNA、UPAR mRNA(P<0.05)和UPA、UPAR蛋白(P<0.01)均差异有显著性;在每组中UPA与UPAR蛋白表达水平之间均存在良好的相关性(P<0.01)。结论UPA和UPAR的基因和蛋白表达水平可随鼻咽癌的发生、发展、浸润转移而发生改变。UPA和UPAR的高表达在鼻咽癌浸润转移中起重要作用,是鼻咽癌预后不良的表现之一。
Objective To investigate the relationship between UPA, UPAR gene and protein expression and the occurrence, development, invasion and metastasis of nasopharyngeal carcinoma (NPC). Methods In situ hybridization and flow cytometry were used to detect nasopharyngeal chronic inflammation in 20 patients, 20 cases of nasopharyngeal carcinoma without metastasis, nasopharyngeal carcinoma in patients with cervical lymph node metastasis in 20 cases and nasopharyngeal carcinoma lymph nodes and distant Twenty patients with organ metastases and other four groups of patients with UPA, UPAR mRNA and protein expression levels. Results The expression levels of UPA, UPAR mRNA, UPA and UPAR in all groups were in the following order: distant metastasis of nasopharyngeal carcinoma, lymph node metastasis of nasopharyngeal carcinoma, no metastasis of nasopharyngeal carcinoma and chronic nasopharyngeal inflammation group. UPA mRNA, UPAR mRNA (P <0.05) and UPA, UPAR protein (P <0.01) were significantly different among groups. There was a good correlation between UPA and UPAR expression in each group P <0.01). Conclusion The gene and protein expression levels of UPA and UPAR may be changed with the occurrence, development, invasion and metastasis of NPC. The high expression of UPA and UPAR plays an important role in the invasion and metastasis of nasopharyngeal carcinoma, which is one of the poor prognosis of nasopharyngeal carcinoma.
