目的采用HPLC法测定不同生长年限杜仲皮中松脂醇二葡萄糖苷的量。方法色谱柱为Agilent Zorb-ax Eclipse XDB-C18(250 mm×4.6 mm,5μm),流动相为甲醇-水(25∶75),检测波长为277 nm,体积流量1 mL/min,柱温25℃。结果13、22年生杜仲内皮松脂醇二葡萄糖苷量相对较高;13年生杜仲栓皮松脂醇二葡萄糖苷量出现最低点。从杜仲皮中松脂醇二葡萄糖苷总量考虑,22年为最佳采收期。结论本实验建立的杜仲皮松脂醇二葡萄糖苷量测定方法,重现性好,结果准确可靠,为杜仲皮质量控制提供了参考。通过对不同生长年限杜仲皮松脂醇二葡萄糖苷量的研究,为杜仲的合理采收提供了科学依据。
OBJECTIVE To determine the amount of pinoresinol diglucoside in Eucommia ulmoides bark with different growth periods by HPLC. Methods The column was Agilent Zorb-ax Eclipse XDB-C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was methanol-water (25:75), the detection wavelength was 277 nm, the volume flow rate was 1 mL / ℃. Results The amount of endocortinol diglucoside was relatively high in 13th and 22th year, and the lowest amount appeared in 13th year. Considering the total amount of pinoresinol diglucoside in Eucommia bark, 22 years was the best harvesting period. Conclusion The method for the determination of pinoresinol diglucoside in Eucommia bark was established with good reproducibility, accurate and reliable results and provided a reference for the quality control of Eucommia bark. The study on the amount of pinoresinol diglucoside in Eucommia bark of different growing years provided the scientific basis for the reasonable harvest of Eucommia bark.
