本文研究了维甲酸(RA)对体外培养的新生大鼠皮肤上皮基底细胞DNA和蛋白质合成的影响。RA(16 μmol/L)明显降低基底细胞~3H-dT掺入;显著增加~3H-亮氨酸和~3H-组氨酸掺入细胞蛋白质。经聚丙烯酰胺凝胶电泳与双相电泳辅以放射自显影术,证实有5种分子量不同的蛋白质受RA诱导合成,其中3种蛋白质为细胞表面膜蛋白。用~3H-甘露糖标记细胞糖蛋白糖链,发现RA可促进~3H-甘露糖的掺入,而且细胞表面糖蛋白中~3H-甘露糖掺入量比对照组增加43%。结果提示RA抑制上皮基底细胞增殖,诱导细胞合成蛋白质,主要为蛋白质的转换率增强,以及增加糖蛋白糖链合成。
This article studies the effect of retinoic acid (RA) on DNA and protein synthesis in the basal cells of the skin of neonatal rats cultured in vitro. RA (16 μmol / L) significantly reduced 3H-dT incorporation in basal cells; significantly increased 3H-leucine and 3H-histidine incorporation into cellular proteins. Polyacrylamide gel electrophoresis and two-phase electrophoresis supplemented with autoradiography confirmed five kinds of proteins with different molecular weights induced by RA, of which three were cell surface membrane proteins. 3H-Mannose labeling of cellular glycoprotein oligosaccharides showed that RA promoted incorporation of 3H-mannose and incorporation of ~ 3H-mannose into cell surface glycoproteins increased by 43% over the control. The results suggest that RA inhibits proliferation of epithelial basal cells and induces cellular synthesis of proteins, primarily resulting in increased protein turnover and increased glycan synthesis.
