[目的]观察芪竹方对人胃腺癌MGC-803细胞Caspase-3及端粒酶增殖与凋亡的影响,探讨中药复方在抗肿瘤治疗中的作用机制。[方法]选用芪竹方500μg/ml在4、8、12、16 h 4个不同时间点及125、250、500μg/ml3个不同浓度在24 h作用人胃腺癌细胞MGC-803,分别采用Caspase-3分光光度法及TRAP银染法端粒酶活性检测法检测Caspase-3和端粒酶在人胃腺癌细胞MGC-803中的表达。[结果]芪竹方500μg/ml在16 h时可明显提高人胃腺癌细胞MGC-803中Caspase-3的活化程度,在250、500μg/ml作用24 h时可明显降低人胃腺癌细胞MGC-803端粒酶的活性。[结论]芪竹方可能通过介导人胃腺癌MGC-803细胞中Caspase-3的增殖及诱导端粒酶凋亡等多途径多靶点而发挥作用。
[Objective] To observe the effect of Qizhu Recipe on the proliferation and apoptosis of Caspase-3 and telomerase in human gastric adenocarcinoma MGC-803 cells and to explore the mechanism of Chinese herbal compound in anti-tumor therapy. [Method] The human gastric adenocarcinoma cell line MGC-803 was treated with 500g / ml of Qizhu 500ml at different time points of 4, 8, 12, 16 h and 125, 250 and 500μg / ml at different concentrations for 24 h. Caspase -3 spectrophotometry and TRAP silver staining were used to detect the expression of Caspase-3 and telomerase in human gastric adenocarcinoma cell line MGC-803. [Result] QHD 500μg / ml could significantly increase the activation of Caspase-3 in human gastric adenocarcinoma MGC-803 cells at the dose of 250 and 500μg / ml for 24 h, 803 telomerase activity. [Conclusion] Qizhu Recipe may play a role in multi-target and multi-target through the proliferation of Caspase-3 in human gastric adenocarcinoma MGC-803 cells and the induction of telomerase apoptosis.
