[Objective] To observe the key parameters of enzymatic extraction that impact extraction of indirubin from Baphicacanthus cusia (Nees) Bremek leaves. [Method] The content of indirubin was determined using HPLC. Effects of enzyme dosage, hydrolysis pH and hydrolysis time on extraction rate of indirubin were investigated with the aim of optimizing its extraction technology. [Result] The optimal enzyme-assisted extraction conditions of indirubin from B. cusia leaves were the combination of enzyme dosage 25 U/g, hydrolysis pH 4.8 and hydrolysis time 6 h. Under the above conditions, the Z value of indirubin was 7.02%. [Conclusion] Enzyme-assisted extraction method is suitable for extraction of indirubin from B. cusia leaves.
