目的观察脑出血后抑郁模型大鼠脑内脑源性神经营养因子(BDNF)的表达,研究益肾调气法中药颐脑解郁方治疗脑出血后抑郁的机理。方法以旷场法(open-field)进行行为学评分,选择得分相近的32只大鼠,随机分为正常组、模型组、中药颐脑解郁方组和百忧解组共4组,每组8只。采用免疫组织化学方法观察各组大鼠脑内BDNF的表达变化。结果模型组海马CA1、CA3区BDNF阳性细胞面密度、光密度,CA2区光密度较正常组显著减少(P<0.01),百忧解组CA1区BDNF阳性细胞面密度和CA3区面密度、光密度较模型组显著增加(P<0.01);中药组CA3区BDNF阳性细胞面密度、光密度较模型组增加(P<0.01)。与正常组相比,模型组扣带皮质面密度、光密度及下丘脑面密度显著降低(P<0.01);中药组扣带皮质光密度及下丘脑阳性细胞面密度较模型组显著增加(P<0.01)。结论益肾调气法中药颐脑解郁方能通过增加脑出血后抑郁模型大鼠BDNF的表达,促进神经元的修复与再生,改善抑郁症状。
Objective To observe the expression of brain-derived neurotrophic factor (BDNF) in the rat model of intracerebral hemorrhage depression and to study the mechanism of Yi-na-qi-regulating traditional Chinese medicine (YNA) on the depression after cerebral hemorrhage. Methods Behavioral score was determined by open-field method. Thirty-two rats with similar scores were randomly divided into normal group, model group, Chinese medicine Yinao Jieyufang group and Prozac group, 4 rats in each group only. Immunohistochemistry was used to observe the changes of BDNF expression in rat brain. Results The surface density, optical density and optical density of BDNF positive cells in CA1 and CA3 area of model group were significantly decreased compared with those in normal group (P <0.01). The average density of BDNF positive cells and CA3 area density in CA1 area of Prozac group were Compared with model group, the density and density of BDNF positive cells in CA3 area of TCM group increased significantly (P <0.01). Compared with the normal group, the cingulate cortical area density, optical density and hypothalamic area density in the model group were significantly decreased (P <0.01); the cingulate cortex optical density and the hypothalamic area density in the model group were significantly increased (P <0.01). Conclusion Yishen Tiaochang Chinese herbal medicine Yinao Jieyu can increase the expression of BDNF in the rat model of depression after intracerebral hemorrhage, promote the repair and regeneration of neurons, and improve the depressive symptoms.
