为探讨脂蛋白 ( a)对巨噬细胞释放一氧化氮的影响 ,进一步了解脂蛋白 ( a)在动脉粥样硬化发生发展中的作用 ,将脂蛋白 ( a) (终浓度为 2 .5~ 40 mg/L )与鼠腹腔巨噬细胞共孵 ,检测上清液中乳酸脱氢酶活性、一氧化氮含量和细胞一氧化氮合酶活性。结果发现 ,在基础及脂多糖诱导状态下 ,细胞上清液中一氧化氮含量与脂蛋白 ( a)呈显著剂量依赖性降低 ,并呈时间效应。而细胞一氧化氮合酶活性则不受脂蛋白 ( a)刺激的影响。结果提示脂蛋白 ( a)对巨噬细胞释放 NO有明显抑制作用
In order to investigate the effect of lipoprotein (a) on the release of nitric oxide by macrophages and to further understand the role of lipoprotein (a) in the development of atherosclerosis, lipoprotein (a) (final concentration of 2.5 ~ 40 mg / L) were incubated with murine peritoneal macrophages to detect the activity of lactate dehydrogenase, nitric oxide and nitric oxide synthase in the supernatant. The results showed that under the basal and lipopolysaccharide-induced conditions, the content of nitric oxide in the cell supernatant decreased in a dose-dependent manner with lipoprotein (a) in a time-dependent manner. While nitric oxide synthase activity was not affected by lipoprotein (a) stimulation. The results suggest that lipoprotein (a) significantly inhibits NO release from macrophages
