目的:观察高糖环境下豚鼠膀胱Cajal样细胞形态学变化。方法:应用酶解法分离培养豚鼠膀胱Cajal样细胞,分为无糖组、正常对照组(葡萄糖浓度5mmol/L)、高糖组(葡萄糖浓度分别为15、30、60mmol/L),通过光学倒置显微镜及c-kit抗体染色后激光扫描共聚焦显微镜观察细胞形态。结果:葡萄糖浓度分别为30、60mmol/L组较无糖组、正常对照组(5mmol/L)及15mmol/L组细胞数量减少,差异有统计学意义(P<0.05),蛋白标记后显示蛋白染色部位减少,有核着色迹象。结论:高糖环境可导致Cajal样细胞的形态异常,数量减少,可能引起其功能学的改变,提示其可能是糖尿病膀胱病变的的影响因素。
Objective: To observe the morphological changes of Cajal-like cells in guinea pig bladder under high glucose. Methods: The bladder Cajal-like cells of guinea pigs were isolated and cultured by enzymolysis. The cells were divided into glucose-free group, normal control group (glucose concentration 5mmol / L), high glucose group (glucose concentration 15,30,60mmol / L) Cell morphology was observed by laser scanning confocal microscope after staining with c-kit antibody and microscope. Results: Compared with control group (5mmol / L) and 15mmol / L group, the glucose concentrations in 30,60mmol / L group were significantly decreased (P <0.05) Dyeing site reduction, nuclear coloring signs. CONCLUSION: High glucose environment can cause Cajal-like cells to become morphologically abnormal and decrease in number, which may lead to functional changes. This suggests that Cajal-like cells may be the influencing factors of diabetic bladder disease.
