目的:探讨SLP-2siRNA对裸鼠胃癌移植瘤细胞增殖及凋亡的影响.方法:设计合成化学修饰的SLP-2siRNA,将胃癌细胞SGC-7901接种于裸鼠皮下建立裸鼠胃癌移植瘤模型,随机分为转染SLP-2siRNA组、阴性对照组和空白对照组,于肿瘤局部分别多点多次注射化学修饰SLP-2siRNA、阴性对照siRNA及生理盐水.检测3组移植瘤细胞增殖及凋亡状况,RT-PCR及免疫组织化学技术检测移植瘤细胞内SLP-2mRNA及蛋白的表达.结果:与两对照组相比,注射化学修饰的SLP-2siRNA后,裸鼠胃癌移植瘤细胞生长速度减慢、移植瘤体积减小(P=0.009,P=0.003).抑制瘤率分别为26.74%和30.15%,细胞凋亡无明显变化(P>0.05),肿瘤细胞内SLP-2mRNA及蛋白的表达量降低.结论:SLP-2siRNA可抑制人胃癌裸鼠移植瘤的生长,但对细胞凋亡无影响,提示SLP-2基因参与促进胃癌细胞增殖.
Objective: To investigate the effect of SLP-2 siRNA on the proliferation and apoptosis of gastric cancer xenografts in nude mice.Methods: The gastric cancer SGC-7901 was inoculated subcutaneously in nude mice to establish a model of gastric cancer xenografts, Randomly divided into SLP-2siRNA transfected group, negative control group and blank control group, multiple local injection of SLP-2siRNA, negative control siRNA and normal saline were injected into the tumor site to detect the proliferation and apoptosis of the three groups RT-PCR and immunohistochemistry were used to detect the expression of SLP-2mRNA and protein in the transplanted tumor cells.Results: Compared with the two control groups, the growth of the transplanted gastric cancer cells in nude mice decreased after injection of chemically modified SLP-2siRNA (P = 0.009, P = 0.003). The inhibitory rate of tumor was 26.74% and 30.15% respectively, there was no significant change in apoptosis (P> 0.05), the expression of SLP-2 mRNA and protein in tumor cells Conclusion: SLP-2siRNA can inhibit the growth of human gastric cancer xenografts in nude mice, but has no effect on cell apoptosis, suggesting that SLP-2 gene is involved in promoting the proliferation of gastric cancer cells.
