目的探讨二黄方对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)大鼠不同时间点血浆IFN-γ、IL-4及Th1/Th2(IFN-γ/IL-4)平衡的影响。方法用髓鞘碱性蛋白(myelin basic protein,MBP)免疫Lewis大鼠诱导EAE模型,采用随机数字表法分为模型组、激素组、中药组及正常对照组,分别在免疫后第7天、14天和28天采集血样,检测血浆细胞因子水平。结果免疫后第28天,模型组和激素组IL-4水平均显著下降,低于正常组(P<0.01),中药组IL-4水平升高,与正常组比较差异无统计学意义(P>0.05);免疫后第28天,模型组和激素组Th1/Th2水平均显著升高,与正常组比较差异有统计学意义(P<0.01),中药组Th1/Th2水平降低,与正常组比较差异无统计学意义(P>0.05)。结论二黄方可减轻EAE疾病严重程度,可能通过上调IL-4水平,下调Th1/Th2比值发挥对EAE的治疗作用。
Objective To investigate the effect of Erhuang Fang on the balance of plasma IFN-γ, IL-4 and Th1/Th2 (IFN-γ/IL-4) at different time points in experimental autoimmune encephalomyelitis (EAE) rats. . Methods The Lewis-induced EAE model was immunized with myelin basic protein (MBP) and divided into model group, hormone group, Chinese medicine group and normal control group by random number table method, respectively, on the 7th day after immunization. Blood samples were collected on days 14 and 28 to measure plasma cytokine levels. Results On the 28th day after immunization, the levels of IL-4 in the model group and the hormone group decreased significantly compared with the normal group (P<0.01). The levels of IL-4 in the Chinese medicine group increased, and there was no significant difference compared with the normal group (P). >0.05); On the 28th day after immunization, the Th1/Th2 levels in the model group and the hormone group increased significantly (P<0.01), and the Th1/Th2 levels in the Chinese medicine group decreased compared with the normal group. There was no significant difference between the two groups (P>0.05). Conclusion “Erhuang Recipe” can reduce the severity of EAE disease and may play a therapeutic role in EAE by up-regulating IL-4 level and down-regulating Th1/Th2 ratio.
