目的研究标本的凝血及溶血对HIV/AIDS患者外周血CD4+、CD8+T淋巴细胞计数的影响。方法选取HIV/AIDS患者30例,以抗凝全血、部分凝血、部分溶血血样分组,3组同时染色、裂解红细胞,流式细胞术检测CD4+、CD8+T淋巴细胞计数,对比3组结果差异。结果抗凝全血组CD4+T淋巴细胞数平均为(454.60±181.20)个/μl,部分凝血组为(294.33±132.75)个/μl,2组差异有统计学意义(P<0.05),部分凝血组CD8+、CD3+T淋巴细胞计数也明显减少,组间比较差异均有统计学意义(P<0.05);部分溶血组CD4+T淋巴细胞数平均为(505.47±182.05)个/μl,与抗凝全血组比较差异具有统计学意义(P<0.05),溶血组CD8+T淋巴细胞计数减少,2组比较差异具有统计学意义(P<0.05),但CD3+T淋巴细胞计数2组间比较差异无统计学意义(P>0.05)。结论血液凝固使CD4+、CD8+T淋巴细胞测定值减少,而溶血会增加CD4+T淋巴细胞测定值,减少CD8+T淋巴细胞计数。因此,检验人员应该加强责任心,规范化操作,采集合格的血液标本。
Objective To study the influence of coagulation and hemolysis on the counts of CD4 + and CD8 + T lymphocytes in peripheral blood of HIV / AIDS patients. Methods Thirty patients with HIV / AIDS were enrolled in this study. The anticoagulant whole blood, partial blood coagulation and partial hemolytic blood samples were divided into three groups. The numbers of CD4 + and CD8 + T lymphocytes were detected by flow cytometry. . Results The average number of CD4 + T lymphocytes in anticoagulant whole blood group was (454.60 ± 181.20) / μl, and in partial coagulation group was (294.33 ± 132.75) / μl, with significant difference between the two groups (P <0.05) The numbers of CD8 + and CD3 + T lymphocytes in the coagulation group were also significantly decreased (P <0.05). The average number of CD4 + T lymphocytes in the partial hemolysis group was (505.47 ± 182.05) / μl, (P <0.05). The counts of CD8 + T lymphocytes in hemolysis group were decreased, the differences between the two groups were statistically significant (P <0.05), but the counts of CD3 + T lymphocytes in two groups were statistically significant There was no significant difference between the two groups (P> 0.05). Conclusion Blood coagulation decreases the measured values of CD4 + and CD8 + T lymphocytes, whereas hemolysis increases the measured values of CD4 + T lymphocytes and decreases the counts of CD8 + T lymphocytes. Therefore, the inspectors should strengthen their sense of responsibility, standardize operations, collecting qualified blood specimens.
