目的建立反相高效液相色谱法测定心脉通颗粒中葛根素含量的方法。方法采用BOS Hypersil C18 ODS2(150mm×4.6mm,5um)色谱柱,甲醇-水(25:75)为流动相,流速为1mlmin-1,检测波长为250nm。按外标法进行检测。结果葛根素在浓度为4.08μgml-1~40.8μgml-1范围内线性关系良好,r=0.9999,重复性实验RSD为0.10%,样品加样平均回收率为103.07%(n=9),RSD为1.04%。结论该方法简便准确、重复性好、专属性强,可作为心脉通颗粒的含量测定有效方法。
Objective To establish a method for the determination of puerarin in Xinmaitong Granules by reversed-phase high performance liquid chromatography. Methods A BOS Hypersil C18 ODS2 column (150mm × 4.6mm, 5um) was used. The mobile phase was methanol-water (25:75). The flow rate was 1ml min-1 and the detection wavelength was 250nm. According to external standard test. Results Puerarin had a good linearity in the range of 4.08μgml-1 to 40.8μgml-1 with r = 0.9999, RSD of 0.10%, and average recovery of 103.07% (n = 9) 1.04%. Conclusion The method is simple, accurate, reproducible and specific. It can be used as an effective method for the determination of Xintuitong granules.
