观察抗癫痫中药草果知母汤在阻断戊四唑点燃模型形成过程中 ,对兴奋性氨基酸类神经递质Glu的受体NMDA结合力及其NMDAR1mRNA的影响。以NMDA的竞争性拮抗剂MK 80 1为对照 ,用配基受体法、原位杂交法检测相应指标的变化。结果显示 :与模型组相比 ,中药组大鼠海马NMDAR1mRNA的含量明显降低 ,差别有显著性意义 (P <0 0 5) ,MK 80 1组无明显变化 ;与模型组相比 ,中药组NMDA受体结合力明显降低 ,但差别无显著性意义 ,MK 80 1组皮层NMDA结合力明显降低 ,差别有极显著性意义 (P <0 0 1)。说明中药草果知母汤主要通过降低NMDAR1mRNA的表达来降低NMDA受体的含量。
Observed the effect of anti-epileptic herb Caogu Zhimu Decoction on blocking the pentylenetetrazole kindling model, the NMDA binding ability and NMDAR1 mRNA of the Glu receptor of the excitatory amino acid neurotransmitter. MK 80 1 , a competitive antagonist of NMDA, was used as a control, and changes in the corresponding indicators were detected using a ligand receptor assay and in situ hybridization. The results showed that compared with the model group, the content of NMDAR1 mRNA in the hippocampus of the TCM group was significantly decreased (P < 0.05), and there was no significant change in the MK 80 1 group. Compared with the model group, the NMDA of the TCM group The receptor binding capacity was significantly reduced, but the difference was not significant. The NMDA binding capacity of the cortex of the MK 80 1 group was significantly reduced, and the difference was extremely significant (P < 0.01). The results showed that traditional Chinese medicine Caoguo Zhimu Decoction can reduce the content of NMDA receptor mainly by reducing the expression of NMDAR1 mRNA.
