目的:构建木通属植物的DNA指纹图谱,探讨利用扩增片段长度多态性(AFLP)分子标记在木通属植物遗传多样性、亲缘关系及种质鉴别上的可行性。方法:采用AFLP技术,随机选用8对EcoRⅠ/MseⅠ引物组合构建样品DNA指纹图谱。结果:从64对引物中筛选出带型丰富、多态性较高的8对引物,共检测到977条多态性条带,多态性比率达92.61%。遗传相似系数以及UPGMA聚类分析结果表明:在遗传相似系数0.49处,12个供试样品分为2类:木通为一类,白木通和所有三叶木通聚为一类;此与基于形态学方面的分类结果一致。结论:AFLP技术适用于木通属植物的亲缘关系及种质鉴别分析。
OBJECTIVE: To construct the DNA fingerprinting of Genus Wood and to explore the feasibility of using AFLP molecular markers to identify the genetic diversity, genetic relationship and germplasm identification of Genus Wood. Methods: AFLP technique was used to randomly select 8 pairs of EcoRⅠ / MseⅠ primers to construct DNA fingerprinting. Results: Sixty pairs of primers with rich polymorphism and high polymorphism were screened out from 64 pairs of primers. A total of 977 polymorphic bands were detected with a polymorphism rate of 92.61%. Genetic similarity coefficient and UPGMA clustering analysis showed that 12 genetic samples were divided into two categories at the genetic similarity coefficient of 0.49: The results of the classification of the same school. Conclusion: The AFLP technique is suitable for the genetic relationship and germplasm identification of the genus.
