目的:检测垂体瘤转化基因(PTTG)在慢性粒细胞白血病(CML)患者中不同临床阶段的基因表达,为阐明CML演变的可能机制提供依据。方法:采用RT-PCR方法检测34例CML患者(慢性期16例,加速期11例,急变期7例)及10例对照者骨髓单个核细胞(BMMNC)中PTTG和c-myc基因的表达。结果:10例对照者中未检测到PTTG和c-myc基因的表达,34例CML患者中PTTG和c-myc基因在慢性期、加速期、急性变期的表达明显高于对照者(P<0.05),PTTG在加速期和急变期的表达升高,但差异无统计学意义(P>0.05)。c-myc基因在急变期的表达明显升高(P<0.05)。结论:PTTG和c-myc基因的过度表达可能和CML的发生发展有关。
Objective: To detect the gene expression of pituitary tumor transforming gene (PTTG) in different clinical stages of patients with chronic myeloid leukemia (CML), and to provide basis for elucidating the possible mechanism of CML evolution. Methods: The expressions of PTTG and c-myc in bone marrow mononuclear cells (BMMNC) were detected by RT-PCR in 34 cases of CML patients (16 chronic, 11 accelerated, 7 blast crisis) and 10 controls. Results: PTTG and c-myc were not detected in 10 controls. The expression of PTTG and c-myc in chronic phase, accelerated phase and acute phase in 34 CML patients was significantly higher than that in controls (P < 0.05). The expression of PTTG in accelerated phase and blastic phase increased, but the difference was not statistically significant (P> 0.05). The expression of c-myc gene in blastic phase was significantly increased (P <0.05). Conclusion: The overexpression of PTTG and c-myc may be related to the development of CML.
