目的:研究热毒宁注射液雾化吸入的刺激性。方法:健康SD大鼠12只,随机分为生理盐水组、热毒宁低浓度组、热毒宁高浓度组,每组4只,各组每日给药2次,每次雾化吸入20min,连续给药7d,观察大鼠行为变化,于第8天取口腔黏膜、鼻中隔黏膜、气管、左肺,制备石蜡切片,HE染色,观察形态学结构变化。同时,右肺做肺泡灌洗术,取灌洗液离心并测定蛋白浓度及乳酸脱氢酶(LDH)活力。结果:大鼠有不同程度的行为反应,给药组有鼻腔出血和眼部出血的现象。切片观察可知热毒宁对黏膜有一定刺激性,且热毒宁高浓度组刺激性大于热毒宁低浓度组。3组肺泡灌洗液蛋白浓度均无显著性差异,而LDH活力随热毒宁浓度的增大而显著增大(P<0.05)。结论:热毒宁雾化吸入给药方式的刺激性较明显,临床应用须谨慎。
OBJECTIVE: To study the irritation induced by atomization inhalation of Renduining injection. Methods: Twelve healthy SD rats were randomly divided into normal saline group, low dose of Renduining group and high dose of Renduining group, with 4 rats in each group. Each group was given 2 times a day for 20 minutes The rats were sacrificed for 7 days. The behavior of the rats was observed. On the 8th day, oral mucosa, mucosa of the nasal septum, trachea and left lung were prepared and paraffin sections were made. HE staining was performed to observe the morphological changes. At the same time, the right lung was treated with alveolar lavage and the perfusate was centrifuged and the protein concentration and lactate dehydrogenase (LDH) activity measured. Results: Rats had different degrees of behavioral response. The treatment group had nasal haemorrhage and ocular hemorrhage. Slice observation shows that verapamil has some irritation to the mucous membrane, and the high concentration of heat toxic rather irritating than the low concentration of heat toxicity group. There was no significant difference in the concentration of alveolar lavage fluid between the three groups, but LDH activity increased significantly with the increase of the concentration of Hyperthermus (P <0.05). CONCLUSION: The treatment of nebulized nebulized inhalation is more irritating and clinical application should be cautious.
