目的:探讨缺氧缺血性脑损伤大鼠脑组织中SDF-1与NSCs各时间点表达情况和作用。方法:72只7日龄SD大鼠随机分为对照组36只和模型组36只。模型组参照Rice法单侧结扎新生大鼠左颈总动脉复制HIBD动物模型,对照组只切开颈部正中皮肤,分离但不结扎左侧颈总动脉。处死前2小时腹腔注射BrdU,对照组和模型组分别在3小时,1天,3天,7天,10天,14天6个时间点处死6只,采用HE染色观察脑组织变化情况,免疫组织化学染色观察海马区SDF-1和BrdU的表达情况。结果:HIBD组SDF-1在1天时表达开始明显升高,3天达到高峰,到7天时略有点降低,组内比较差异性显著,有统计学意义(p<0.05),组间各时间点差异显著,有统计学意义(p<0.05),对照组各时间点差异无显著,无统计学意义(p>0.05)。BrdU表达在1天表达很少,在3天表达增高,7天表达开始下降,14天表达更少。IODBrdU阳性细胞/IODSDF-1阳性细胞的均值比在3天左右最大。结论:SDF-1可能趋化内源性神经干细胞修复脑损伤,并且在3天左右时能够更大程度修复脑损伤。
Objective: To investigate the expression of SDF-1 and NSCs in the brain tissue of rats with hypoxic-ischemic brain damage at different time points. Methods: Seventy-two 7-day-old SD rats were randomly divided into control group (36) and model group (36). Model group reference Rice method unilateral ligation of neonatal rats left common carotid artery replication HIBD animal model, the control group only cut open the middle of the neck skin, but without the left common carotid artery ligation. BrdU was intraperitoneally injected 2 hours before sacrifice, and 6 rats were killed at 6, 3, 7, 10, 14 days respectively in the control group and the model group. HE staining was used to observe the changes of brain tissue, Histochemical staining was used to observe the expression of SDF-1 and BrdU in hippocampus. Results: The expression of SDF-1 in HIBD group was significantly increased at 1 day and peaked at 3 days, slightly decreased at 7 days. There was significant difference between the two groups (p <0.05) There was significant difference between the two groups (p <0.05). There was no significant difference in the control group at each time point (p> 0.05). BrdU expression was low on day 1, increased on day 3, decreased on day 7, and decreased on day 14. The mean ratio of IODBrdU positive cells / IODSDF-1 positive cells was the largest in about 3 days. Conclusion: SDF-1 may chemotaxic the endogenous neural stem cells to repair brain injury, and repair the brain injury to a greater extent in about 3 days.
