目的建立超高效液相色谱法同时测定黄芩中6种黄酮类成分的含量。方法采用Waters Acquity UPLC BEH C18柱(2.1 mm×100 mm,1.8μm),流动相:乙腈(A)-体积分数为0.05%的磷酸水溶液(B)梯度洗脱[0 min:A-B(体积比为30∶70),5 min:A-B(体积比为50∶50);9 min:A-B(体积比为60∶40)],流速:0.4 m L·min-1,检测波长:220 nm,柱温:30℃。结果 6个黄酮(黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、白杨素和千层纸素A)的色谱峰面积与质量浓度呈良好的线性关系,线性分别为24.4~292.1(r=0.999 5)、8.5~102.4(r=0.999 7)、6.8~82.0(r=0.999 5)、3.1~37.4(r=0.999 5)、0.6~6.7(r=0.999 5)、1.2~14.8 mg·L-1(r=0.999 6);平均回收率(n=6)分别为99.9%、100.8%、97.7%、100.1%、97.6%、98.9%。结论本方法可被用于黄芩中黄酮类成分的定量分析。
OBJECTIVE To establish a method for the simultaneous determination of six flavonoids in Scutellaria baicalensis Georgi by ultra performance liquid chromatography. Methods A Waters Acquity UPLC BEH C18 column (2.1 mm × 100 mm, 1.8 μm) was used. The mobile phase consisted of a gradient of acetonitrile (A) - 0.05% phosphoric acid in water 30:70), 5 min: AB (volume ratio of 50:50), 9 min (volume ratio of 60:40), flow rate of 0.4 m L · min-1, detection wavelength of 220 nm, : 30 ° C. Results The chromatograms of 6 flavonoids (baicalin, baicalin, baicalein, wogonin, chrysin, melatonin A) showed a good linear relationship with the mass concentration and their linearities were 24.4 ~ 292.1 (r = 0.999 5, 8.5 ~ 102.4 r = 0.999 7, 6.8 ~ 82.0 r = 0.999 5, 3.1 ~ 37.4 r = 0.999 5, 0.6 ~ 6.7 r = 0.999 5 and 1.2 ~ 14.8 mg · L -1 (r = 0.999 6). The average recoveries (n = 6) were 99.9%, 100.8%, 97.7%, 100.1%, 97.6% and 98.9%, respectively. Conclusion This method can be used for the quantitative analysis of flavonoids in Scutellaria baicalensis Georgi.
