目的:消癌平是传统用于治疗咳嗽和哮喘的萝藦科中药通光藤的提取物,具有抗炎和抗肿瘤作用。通过体内外研究消癌平对人胃癌SGC-7901细胞诱导凋亡作用,而探讨其抗肿瘤活性。方法采用消癌平药物剂量2040mg·mL1作用SGC-7901细胞24和48h。体外实验采用MTT检测法评估药物对细胞生长和细胞活力,流式细胞仪检测药物对SGC-7901细胞凋亡和周期阻滞作用,体内实验采用裸鼠移植瘤模型检测其体内抗肿瘤活性,免疫组化和细胞凋亡TUNEL法检测药物对荷瘤鼠体内肿瘤组织中的凋亡蛋白Caspase-3,Bax和Bcl-2的表达。结果:消癌平可以显著抑制SGC-7901细胞的生长,并存在时间和剂量依赖性性,经过24h药物处理后其IC50约在(38.20±0.27)mg·mL1。流式细胞仪分析消癌平可诱导SGC-7901细胞凋亡和周期阻滞实验显示其可将SGC-7901细胞周期阻滞于G1。体内实验显示,经消癌平治疗组的肿瘤体积和重量都存在显着降低,中剂量组和高剂量组的抑瘤率分别为61.19%和69.07%。免疫组化和细胞凋亡TUNEL法检测显示,消癌平治疗组小鼠肿瘤组织中的Bax和caspase-3蛋白表达量显著增加,而Bcl-2的表达量下降。结论:消癌平在体外和体内对人胃癌SGC-7901细胞均有抗肿瘤活性,并可能通过诱导SGC-7901细胞内凋亡蛋白Bax和caspase-3蛋白的表达这一机制发挥抗肿瘤作用。
Objective: Xiaochangping is a traditional Chinese herbal medicine extract of Rhodotorulae used in the treatment of cough and asthma. It has anti-inflammatory and antitumor effects. To study the anti-tumor activity of Xiaochangping on human gastric cancer SGC-7901 cells in vitro and in vivo. Methods SGC-7901 cells were treated with 2040 mg · mL -1 of HOPE for 24 and 48 hours respectively. In vitro experiments MTT assay was used to evaluate the effects of drugs on cell growth and cell viability. Flow cytometry was used to detect apoptosis and cell cycle arrest of SGC-7901 cells. In vivo, the antitumor activity in vivo was detected by in vivo nude mice model. TUNEL assay was used to detect the expression of Caspase-3, Bax and Bcl-2 in tumor tissues of tumor-bearing mice. Results: Xiaojianping could significantly inhibit the growth of SGC-7901 cells in a time-and dose-dependent manner. The IC50 of SGC-7901 cells was (38.20 ± 0.27) mg · mL1 after 24h drug treatment. Flow cytometry analysis of HCC can induce SGC-7901 cell apoptosis and cycle arrest experiments showed that it can block SGC-7901 cell cycle in G1. In vivo experiments showed that the tumor volume and weight of the treated group were significantly reduced, and that of the middle-dose and high-dose groups were 61.19% and 69.07%, respectively. Immunohistochemistry and apoptosis TUNEL assay showed that the expression of Bax and caspase-3 protein in tumor cells of Xiaochangping treatment group increased significantly, while the expression of Bcl-2 decreased. Conclusion: Xiaofuping has antitumor activity on SGC-7901 cells both in vitro and in vivo and may exert anti-tumor effect by inducing the expression of Bax and caspase-3 protein in SGC-7901 cells.
