目的:建立降糖中药制剂中违法添加格列吡嗪的检测方法。方法:采用高效液相色谱结合二极管阵列紫外光谱检测,色谱柱:Diamonsil C18柱,流动相:甲醇-0.038 mol·L-1磷酸铵缓冲液(pH 3.5)(51:49),流速1.0 ml·min-1,检测波长:275 nm。结果:格列吡嗪在进样量0.26-5.12μg范围内呈良好线性关系,r=0.999 8,平均回收率为97.8%(n=5),RSD 1.6%, 检测限为1 ng。结论:本法具良好的灵敏度和专属性,能对降糖中药制剂中违法添加的格列吡嗪进行准确的定性和定量检测。
Objective: To establish a method for detecting the illegal addition of glipizide in the traditional Chinese medicine preparation for reducing blood sugar. Method: High performance liquid chromatography combined with diode array UV spectroscopy, column: Diamonsil C18 column, mobile phase: methanol-0.038 mol·L-1 ammonium phosphate buffer (pH 3.5) (51:49), Flow rate 1.0 ml·min-1, detection wavelength: 275 nm. Results: Glipizide showed good linearity in the range of 0.26-5.12μg injection volume, r=0.999 8, the average recovery was 97.8% (n=5), RSD 1.6% The detection limit is 1 ng. Conclusion: This method has good sensitivity and specificity, and can accurately and qualitatively detect glipizide added illegally in the traditional Chinese medicine for hypoglycemic agents.
