目的:探讨LASS2在鼻咽癌(nasopharyngeal carcinoma,NPC)的发生与发展中的作用。方法:收集2011年6月至2014年12月佛山市顺德区中医院治疗并随访的Ⅰ~Ⅳ期鼻咽癌病例127例,其中鼻咽癌Ⅰ期12例(A组),鼻咽癌Ⅱ期26例(B组),鼻咽癌Ⅲ期52例(C组),鼻咽癌Ⅳ期37例(D组),以及慢性鼻咽炎病例49例(E组)。采用免疫组织化学方法检测LASS2蛋白在鼻咽活检组织中的表达。并收集鼻咽癌各组的相关临床资料,分析LASS2表达与各临床因素之间的相关性。结果:LASS2在鼻咽癌病例中的表达率(A,B,C,D组表达的百分率分别是75%,73.1%,78.8%及81.1%)低于慢性鼻咽炎病例中的表达率(E组表达的百分率为100%),其差别具有统计学意义(P=0.008)。LASS2的表达与肿瘤的复发存在负相关性(P=0.001),但LASS2阳性病例与LASS2阴性病例的无瘤生存期的差别无统计学意义(P>0.05)。结论:LASS2是一种抑癌基因,其蛋白产物表达于非肿瘤性的鼻咽组织中。鼻咽癌发生过程中可能发生了蛋白的丢失,从而促进鼻咽癌的发病。LASS2蛋白的丢失与鼻咽癌复发也存在相关性,LASS2可能成为预测鼻咽癌复发的有临床意义的生物学指标。
Objective: To investigate the role of LASS2 in the pathogenesis and development of nasopharyngeal carcinoma (NPC). Methods: A total of 127 cases of stage Ⅰ ~ Ⅳ nasopharyngeal carcinoma were treated and followed up in Shunde District Hospital of Traditional Chinese Medicine from June 2011 to December 2014. Among them, 12 cases of stage Ⅰ nasopharyngeal carcinoma (group A), nasopharyngeal carcinoma Ⅱ Thirty-six patients (group B), 52 patients with stage Ⅲ nasopharyngeal carcinoma (group C), 37 patients with stage Ⅳ nasopharyngeal carcinoma (group D) and 49 patients with chronic nasopharyngitis (group E). Immunohistochemistry was used to detect the expression of LASS2 protein in nasopharyngeal biopsies. The clinical data of NPC patients were collected and the correlation between LASS2 expression and clinical factors was analyzed. Results: The expression of LASS2 in NPC cases (75%, 73.1%, 78.8% and 81.1% in group A, B, C and D respectively) was lower than that in patients with chronic nasopharyngitis The percentage of group expression was 100%), the difference was statistically significant (P = 0.008). There was a negative correlation between LASS2 expression and tumor recurrence (P = 0.001), but there was no significant difference between LASS2 positive and LASS2 negative (P> 0.05). Conclusion: LASS2 is a tumor suppressor gene whose protein product is expressed in non-neoplastic nasopharyngeal tissues. Nasopharyngeal carcinoma may occur during the loss of protein, thereby promoting the incidence of nasopharyngeal carcinoma. LASS2 protein loss and recurrence of nasopharyngeal carcinoma are also related, LASS2 may be a prediction of nasopharyngeal cancer recurrence of clinically significant biological indicators.
