木贼作为一种重要的中药材,人们常与同属其他植物混用,这直接影响到人们的身体健康。该实验拟从常用DNA条形码(ITS,mat K,rbc L和psb A-trn H)中筛选适合木贼属的核心DNA条形码,并存储正品及其混伪品的参考序列,便于今后查询,确保用药安全。本实验采集了14个样本,并从Gen Bank上获得木贼属121条序列。应用PAUP4.0软件计算K2P遗传距离,通过5个参数评估序列变异,构建NJ系统发育树,最终评价ITS,mat K,rbc L和psb A-trn H序列对木贼属植物的鉴别能力。研究结果表明,14个样本获得100%的DNA提取成功率;psb A-trn H,mat K,rbc L和ITS的扩增效率分别为:100%,96%,92%和52%。rbc L序列存在明显的Barcoding Gap,所构建的NJ系统发育树也能将木贼属植物区分开,鉴别成功率最高,为71.4%。因此,建议把rbc L序列作为木贼属植物的核心DNA条形码。
Equisetum as an important Chinese herbal medicine, people often mix with other plants, which directly affects people’s health. This experiment intends to screen core DNA barcodes for Aristichthys nobilis from commonly used DNA barcodes (ITS, mat K, rbc L and psb A-trn H) and store the reference sequences of genuine and its adulterants for future reference to ensure Medication safety In this experiment, 14 samples were collected and 121 ESTs were obtained from Gen Bank. The genetic distance of K2P was calculated using PAUP4.0 software, and the sequence variation was evaluated by five parameters to construct NJ phylogenetic tree. Finally, the discrimination ability of ITS, mat K, rbc L and psb A-trn H sequences on the genus Gypsophila was evaluated. The results showed that the success rate of 100% DNA extraction from 14 samples was 100%, 96%, 92% and 52% for psb A-trn H, mat K, rbc L and ITS, respectively. The Barcoding Gap existed in the rbc L sequence. The NJ phylogenetic tree also distinguished the Gypsophila species, with the highest success rate of 71.4%. Therefore, it is suggested that the rbc L sequence be used as the core DNA barcode for Equisetum.
