目的:探讨中药复方外敷剂消痹膏对牛Ⅱ型胶原诱发的大鼠关节炎的抗炎及免疫调节作用。方法:将牛Ⅱ型胶原溶液与不完全弗氏佐剂混合配制的乳剂注射于Wistar大鼠诱发免疫性关节炎(CIA)大鼠模型,取造模成功的大鼠随机分为模型组、阳性对照组(扶他林乳胶剂)及消痹膏低、中、高剂量组;另外设注射生理盐水的大鼠为阴性对照组。模型组与阴性对照组于腹部剔毛处外敷黄凡士林,其余各组分别予外敷相应药物。考察各组关节炎指数及抗炎、免疫调节作用。结果:与模型组比较,消痹膏各剂量组给药7 d大鼠足趾容积明显减少(P<0.01或P<0.05),中剂量组给药2 d大鼠关节炎指数明显降低(P<0.05),低、高剂量组给药3 d关节炎指数明显降低(P<0.01或P<0.05);消痹膏高剂量组对CIA大鼠组织病理变化分级的改善疗效显著增强(P<0.01),血清IL-4含量显著升高(P<0.01)、血清TGF-β含量明显降低(P<0.05);中剂量组血清TNF-α含量明显降低(P<0.05)。与阳性对照组比较,消痹膏低剂量组大鼠组织病理变化分级R值显著降低(P<0.01),低、中、高剂量组关节炎指数评分、足趾容积、细胞因子含量差异均无统计学意义。结论:消痹膏有明显的抗炎、免疫调节作用。
OBJECTIVE: To investigate the anti-inflammatory and immunomodulatory effects of Xiaobai Paste, a compound traditional Chinese medicine compound, on arthritis induced by bovine type Ⅱ collagen in rats. Methods: The emulsion prepared by mixing bovine type Ⅱ collagen with incomplete Freund’s adjuvant was injected into the rat model of autoimmune arthritis (CIA) induced by Wistar rats. The successful model rats were randomly divided into model group, positive Control group (Fu Tailin latex) and Xiaobai low, medium and high dose group; another injection of normal saline rat negative control group. The model group and the negative control group applied yellow petrolatum to the abdomen ticking area, while the other groups were given corresponding drugs respectively. The arthritis index and anti-inflammatory and immunomodulatory effects of each group were investigated. Results: Compared with the model group, the volume of the toe in each dose group of Xiaobai was significantly decreased (P <0.01 or P <0.05), and the arthritis index of the middle dose group was significantly decreased (P <0.01 or P <0.05). The high dose group of Xiaobai Gao significantly improved the grading of histopathological changes in CIA rats (P <0.01 or P <0.05) 0.01). The level of serum IL-4 was significantly increased (P <0.01), the level of serum TGF-β was significantly decreased (P <0.05), while the level of serum TNF-α in middle dose group was significantly decreased (P <0.05). Compared with the positive control group, the R value of histopathological changes in the low dose Xiaoxi Pian group was significantly lower (P <0.01), the arthritis index score, the volume of the toe and the content of cytokines in the low, medium and high dose groups were all different Statistical significance. Conclusion: Xiaobai cream has obvious anti-inflammatory and immunomodulatory effects.
