目的探讨核基质结合区结合蛋白1(SATB1)在结直肠癌细胞中的表达及其临床意义。方法以大肠癌细胞株SW480、SW620为研究对象,运用免疫细胞化学分析SATB1在细胞内的表达情况。将142例标本分为癌旁组织、腺瘤组织、大肠癌组织、淋巴结转移癌,运用免疫组织化学方法分析SATB1在这些组织中的表达情况。结果 SATB1在人大肠癌细胞系SW480与SW620细胞中均有较强的表达,并且定位在SW480与SW620细胞的细胞核。SATB1在正常结直肠粘膜、腺瘤、癌及淋巴结转移癌组织中的表达明显不同,差异有统计学意义(χ2=79.653,P<0.05)。SATB1表达与结直肠癌转移高度相关(Z=-2.115,P<0.05),即SATB1表达弱者转移的发生率低,表达强者其转移发生率高。结论SATB1在结直肠癌细胞中高表达,与结直肠癌发生演进及转移存在联系。
Objective To investigate the expression of nuclear matrix binding domain 1 (SATB1) in colorectal cancer cells and its clinical significance. Methods The human colorectal cancer cell lines SW480 and SW620 were used as research objects. The expression of SATB1 in cells was analyzed by immunocytochemistry. 142 cases were divided into adjacent tissue, adenoma, colorectal carcinoma and lymph node metastasis. The expression of SATB1 in these tissues was analyzed by immunohistochemical method. Results SATB1 was strongly expressed in human colorectal cancer cell lines SW480 and SW620, and located in the nucleus of SW480 and SW620 cells. The expression of SATB1 in normal colorectal mucosa, adenoma, cancer and lymph node metastasis was significantly different, the difference was statistically significant (χ2 = 79.653, P <0.05). The expression of SATB1 was highly correlated with the metastasis of colorectal cancer (Z = -2.115, P <0.05), that is, the low incidence of SATB1 metastasis and the strong metastasis of SATB1. Conclusions SATB1 is highly expressed in colorectal cancer cells and is associated with the development and metastasis of colorectal cancer.
