目的:研究白细胞介素-17(IL-17)在人甲状腺乳头状癌组织中的表达,探讨IL-17在甲状腺乳头状癌发生发展中的作用。方法:采用real-time RT-PCR、Western blot、免疫组织化学法检测IL-17在60例甲状腺乳头状癌、10例结节性甲状腺肿和10例桥本氏病组织中的表达,应用统计学方法对其结果进行分析。结果:①甲状腺乳头状癌IL-17 mRNA水平高于结节性甲状腺肿,而低于桥本氏病,其中癌旁组织高于癌组织,P<0.05;②Western blot检测结果显示,IL-17蛋白在结节性甲状腺肿基本不表达,甲状腺乳头状癌癌旁组织表达高于癌组织,但均低于桥本氏病(P<0.05);③免疫组化染色显示IL-17在甲状腺乳头状癌和桥本氏病均有表达,结节性甲状腺肿无表达,且甲状腺癌染成棕色的区域主要位于肿瘤边缘;④临床资料统计显示IL-17的表达与甲状腺乳头状癌患者的年龄、性别、肿瘤个数无关,与肿瘤直径、淋巴结转移以及TNM分期相关。结论:IL-17在甲状腺乳头状癌表达增高,IL-17可能通过影响多种细胞因子以及炎症细胞参与甲状腺癌的发生和发展。
Objective: To investigate the expression of interleukin-17 (IL-17) in human thyroid papillary carcinoma and to explore the role of IL-17 in the development of papillary thyroid carcinoma. Methods: The expression of IL-17 in 60 cases of thyroid papillary carcinoma, 10 cases of nodular goiter and 10 cases of Hashimoto’s disease was detected by real-time RT-PCR, Western blot and immunohistochemistry. Learning methods to analyze the results. Results: ① The level of IL-17 mRNA in papillary thyroid carcinoma was higher than that in nodular goiter and lower than that in Hashimoto’s disease, with paracancer tissue higher than that in cancerous tissues (P <0.05). ②Western blot results showed that IL-17 Protein in nodular goiter basically not expressed, thyroid papillary carcinoma adjacent tissue expression was higher than in cancerous tissue, but were lower than Hashimoto’s disease (P <0.05); ③Immunohistochemical staining showed that IL-17 in thyroid papilla Cancer and Hashimoto’s disease are expressed, no expression of nodular goiter, and thyroid cancer stained brown area mainly located in the edge of the tumor; ④ clinical data statistics show that the expression of IL-17 and thyroid papillary carcinoma of patients age , Gender, the number of tumor has nothing to do with tumor diameter, lymph node metastasis and TNM staging related. Conclusion: The expression of IL-17 is increased in thyroid papillary carcinoma. IL-17 may participate in the occurrence and development of thyroid carcinoma by influencing many cytokines and inflammatory cells.
