目的研究珍珠菜总黄酮(ZE4)对白血病HL-60细胞裸鼠皮下移植瘤的抑制作用。方法成功建立人白血病HL-60细胞皮下移植瘤模型(移植瘤体积约100mm3)后的30只裸鼠均分为五组:A、B、C组分别腹腔注射ZE4 100、200、400mg/kg;D组注射环磷酰胺(CTX)50mg/kg;E组以生理盐水对照;每2天注射一次。21d后测量移植瘤大小,采用免疫组化法检测移植瘤组织中细胞增殖抗原Ki-67和血管内皮细胞膜抗原CD34的表达水平。结果与E组比较,B和C组移植瘤生长明显抑制,抑瘤率分别可达51.32%和46.06%;免疫组化结果显示,B、C组移植瘤中Ki-67和CD34的表达明显少于E组。结论 ZE4对白血病HL-60细胞裸鼠移植瘤具有较明显的抑制作用,可能与其抑制肿瘤细胞增殖和肿瘤新生血管形成相关。
Objective To study the inhibitory effect of ZE4 on subcutaneous xenografts of leukemia HL-60 cells in nude mice. Methods Thirty nude mice were successfully established by subcutaneous xenograft model of human leukemia HL-60 cells (tumor volume about 100mm3) divided into five groups: group A, B and C were intraperitoneally injected with ZE4 100,200,400mg / kg; D group injection of cyclophosphamide (CTX) 50mg / kg; E group with normal saline control; every 2 days injections. The size of the xenografts was measured 21 days later. The expression of Ki-67 and CD34 in the tumor tissue were detected by immunohistochemistry. Results Compared with group E, the growth of transplanted tumor in groups B and C was significantly inhibited and the tumor inhibition rates were 51.32% and 46.06% respectively. Immunohistochemical results showed that the expression of Ki-67 and CD34 in group B and C was significantly decreased In Group E Conclusion ZE4 has a significant inhibitory effect on leukemia HL-60 cells transplanted in nude mice, which may be related to the inhibition of tumor cell proliferation and neovascularization.
