动物组织中3种β-肾上腺受体激动剂残留的高效液相色谱荧光检测法

目的盐酸多巴胺、硫酸沙丁胺醇和盐酸莱克多巴胺是β-肾上腺素受体激动剂,该研究目的是建立一种用于这3种成分的检测方法,以防止人类食用含有该残留成分的畜产品引发食物中毒。方法实验选用SHIMADZU ODS C18反相色谱柱(150 mm×4.6 mm,5μm),以含10%乙腈的0.02 mol/L磷酸二氢钾溶液(p H=2.50)为流动相A、含50%乙腈的0.02 mol/L磷酸二氢钾溶液(p H=2.50)为流动相B,在0~6 min内由A液变为B液进行梯度洗脱。激发波长(λex)为280 nm,发射波长(λem)为309 nm,对3种β-肾上腺素受体激动剂进行分离和检测。结果盐酸多巴胺、硫酸沙丁胺醇和盐酸莱克多巴胺分别在5~200、5~200和10~200 ng范围内呈良好的线性关系,其相关系数R2分别为0.999 3、0.999 9和0.999 2,其标准回收率分别为(101.50±3.42)%、(99.46±2.93)%和(101.83±4.12)%,变异系数为3.37%、2.95%和4.05%。样品的回收率分别为(60.74±1.05)%、(96.30±0.62)%和(91.02±1.28)%,变异系数分别为1.72%、0.64%和1.40%。盐酸多巴胺、硫酸沙丁胺醇和盐酸莱克多巴胺的最低检测限分别为0.1、0.125和5 ng。结论该方法简单,快速,准确,可用于实验室动物性食品中残留药物的常规检测。 Purpose Dopamine hydrochloride, salbutamol sulfate and ractopamine hydrochloride are β-adrenoceptor agonists. The aim of this study was to establish a test method for the three components to prevent human consumption of animal products containing the residual components to trigger food Poisoning. Methods The SHIMADZU ODS C18 reversed-phase column (150 mm × 4.6 mm, 5 μm) was used as the mobile phase A with 0.02 mol / L potassium dihydrogen phosphate solution (p H = 2.50) containing 10% 0.02 mol / L potassium dihydrogen phosphate solution (p H = 2.50) was used as the mobile phase B. The gradient elution was carried out from A to B solution within 0 ~ 6 min. The excitation wavelength (λex) was 280 nm and the emission wavelength (λem) was 309 nm. Three β-adrenoceptor agonists were isolated and detected. Results There was a good linear relationship between dopamine hydrochloride, salbutamol sulfate and ractopamine hydrochloride in the range of 5 ~ 200, 5 ~ 200 and 10 ~ 200 ng respectively. The correlation coefficients R2 were 0.999 3, 0.9999 9 and 0.999 2, respectively. The standard recovery The rates were (101.50 ± 3.42)%, (99.46 ± 2.93)% and (101.83 ± 4.12)%, respectively. Conclusion The method is simple, rapid and accurate and can be used for routine testing of residual drugs in laboratory animal food.
