应用花粉管通道法将大赖草 DNA导入小麦 ,已从受体 761转化后代中选育出稳定遗传的大穗变异系。主要特征为 :( 1 )大穗变异系生育期晚 ,株型高大 ,叶片宽大 ,穗长、穗粒数和单株粒重显著增加 ,同时籽粒长、宽和千粒重明显提高 ;( 2 )大穗变异系除蛋白质含量有显著变化外 ,其胚乳谷蛋白和醇溶蛋白电泳图谱发生了明显变化 ,出现了新的蛋白组分和受体一些带的消失。由此可见 ,大赖草 DNA导入小麦后发生广泛变异 ,可能是外源 DNA片段重组到受体基因组的结果
The pollen tube pathway was used to transfer the DNA of L.sinensis into wheat, and the stable genetic large panicle mutation line was bred from the 761 transformed progeny. The main characteristics are as follows: (1) Large ear variation line has late growth stage, tall plant type, large leaf width, spike length, spike grain number and grain weight per plant increased significantly, meanwhile grain length, width and 1000-grain weight increased obviously; (2) In addition to significant changes in the protein content of the spike variation, the glutenin and prolamin electrophoresis patterns of the endosperm evidently changed, and new protein components and some bands of the receptor disappeared. Thus, a wide range of variation occurs after the introduction of the DNAs of Larvae grass into wheat, possibly as a result of the recombination of exogenous DNA fragments into the recipient genome
