HPLC测定芹菜籽提取物中木犀草素含量

目的建立高效液相色谱法测定芹菜籽提取物中木犀草素的含量。方法采用Apollo C18柱(4.6 mm×250 mm,5μm);流动相为乙腈-0.1%磷酸溶液(30∶70);流速为1.0 mL.min-1;进样量为20μL;检测波长为269 nm;柱温为30℃。结果木犀草素浓度线性范围为0.50~10.0μg.mL-1(r=0.999 8),3个浓度的加样回收率分别为100.2%,100.1%和101.2%,RSD分别为1.02%,1.91%和1.75%。结论本法操作简单、准确、专属性强,可作为芹菜籽提取物质量控制的方法。 Objective To establish a HPLC method for the determination of luteolin in celery seed extract. Methods Apollo C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (30:70), the flow rate was 1.0 mL.min-1 and the injection volume was 20 μL. The detection wavelength was 269 nm The column temperature was 30 ℃. Results The linear range of luteolin was 0.50 ~ 10.0 μg.mL-1 (r = 0.999 8). The recoveries of three concentrations were 100.2%, 100.1% and 101.2%, respectively. The RSDs were 1.02% and 1.91% And 1.75%. Conclusion This method is simple, accurate and specific and can be used as a quality control method for celery seed extract.
