目的 探讨细胞周期转录因子E2F在良性前列腺增生症 (BPH)发病过程中作用。方法 应用免疫组化方法检测E2F、细胞周期蛋白E(cyclinE)、cyclinD1和PCNA在 62例BPH组织和 14例正常前列腺 (NP)组织中的表达 ;应用DNA图像定量分析法测定BPH组织中DNA含量和细胞周期不同时相细胞比率。结果 E2F表达主要位于前列腺上皮细胞 ,BPH中阳性率为 66.1% ,显著高于NP的 2 8.6% (P <0 .0 1)。BPH组织中E2F阳性表达者的G0 /G1期细胞比率较阴性表达者呈降低趋势 (P =0 .0 5 1)、而S期细胞比率、DNA含量和PCNA指数显著高于阴性表达者 (P <0 .0 5 )。BPH组织中E2F表达与cyclinE表达之间密切相关。 结论 E2F异常高表达导致细胞周期紊乱、细胞增殖增加 ,从而促使BPH的发生发展
Objective To investigate the role of cell cycle-associated transcription factor E2F in the pathogenesis of benign prostatic hyperplasia (BPH). Methods The expressions of E2F, cyclinE, cyclinD1 and PCNA were detected by immunohistochemistry in 62 cases of BPH and 14 cases of normal prostate tissue (NP). The DNA content of BPH tissues was determined by DNA image quantitative analysis And the cell cycle at different phases of the cell ratio. Results The positive expression rate of E2F in prostate epithelial cells was 66.1%, which was significantly higher than that of NP (22.6%, P <0.01). The ratio of G0 / G1 phase cells in E2F positive cells in BPH tissue was lower than that in negative cells (P = 0.051), but the S phase cell ratio, DNA content and PCNA index were significantly higher than those in negative cells <0 .0 5). There is a close relationship between E2F expression and cyclinE expression in BPH tissues. Conclusion The abnormally high expression of E2F leads to the disorder of cell cycle and the increase of cell proliferation, which leads to the occurrence and development of BPH
