用Langendorff离体心脏灌流方法证明,山茛菪碱(654-2)可使正常大鼠心肌收缩力减弱,心率减慢,冠腺流量减少;高钙及毒毛旋花子甙K可消除654-2对心肌收缩的抑制作用。654-2与钙拮抗剂异搏停一样,还可对抗大剂量异丙肾上腺素所致的心肌细胞钙超负荷及坏死。以上结果提示,山莨菪碱具有钙拮抗作用。
Langendorff perfusion in vitro showed that anisodamine (654-2) reduced myocardial contractility, slowed heart rate and reduced coronary flow in normal rats. High calcium and toxin K could eliminate 654- 2 on myocardial contraction inhibition. 654-2 is the same as calcium antagonist of verapamil, but also against the large dose of isoprenaline cardiomyocyte calcium overload and necrosis. The above results suggest that anisodamine has a calcium antagonistic effect.
