目的观察复方丹参注射液对严重烫伤大鼠血脑屏障通透性及脑内ZO-1mRNA影响,探讨复方丹参对严重烫伤大鼠血脑屏障损伤的保护作用。方法将大鼠随机分成假烫伤组、烫伤组、复方丹参治疗组。紫外分光光度计检测各组大鼠脑组织内伊文氏蓝的含量,RT-PCR检测严重烫伤大鼠脑内ZO-1 mRNA表达变化。结果大鼠严重烫伤后3~48 h脑内ZO-1mRNA表达下降。脑内伊文氏蓝(EB)含量从烫伤后3h开始升高,在6~12 h显著升高。用复方丹参治疗后,严重烫伤大鼠脑内EB含量降低,以3、6 h下降最明显(P<0.010);脑内ZO-1mRNA表达升高,以烫伤3 h治疗组最为明显。结论复方丹参注射液可防止严重烫伤大鼠脑内ZO-1mRNA的下降,减轻严重烫伤对大鼠血脑屏障的损害。
Objective To observe the effects of Compound Danshen Injection on blood-brain barrier permeability and brain ZO-1 mRNA in severely scalded rats, and to explore the protective effect of compound Danshen on blood-brain barrier injury in severe scald rats. Methods Rats were randomly divided into sham scald group, scald group and compound salvia miltiorrhiza treatment group. Ultraviolet spectrophotometer was used to detect the content of Evans blue in brain tissue of each group. RT-PCR was used to detect the expression of ZO-1 mRNA in brains of severely burned rats. RESULTS: The expression of ZO-1 mRNA in brain was decreased 3 to 48 h after severe scald in rats. The content of Evan’s blue (EB) in the brain increased from 3 h after scald and increased significantly at 6-12 h. After treatment with compound salvia miltiorrhiza, the brain EB content of severely burned rats decreased, and the most significant decrease was at 3 and 6 h (P<0.010). The expression of ZO-1 mRNA in the brain was increased, and the treatment group was most obviously treated with scald for 3 h. Conclusion Compound salvia miltiorrhiza injection can prevent the decrease of ZO-1 mRNA in severely burned rats and reduce the damage of severe scald on blood-brain barrier in rats.
