高效液相色谱法是现代分离分析新技术,在许多学科领域里已得到广泛的应用。目前已有不少分析型、制备型和微量分析型的高效液相色谱仪出现,但这些仪器造价均较贵。本文提出应用由结构简易的气动活塞泵、内径10毫米的不锈钢层析柱、硅胶H作为填充剂以及单波长(254nm)紫外检测器组成的简易高效液相色谱仪。实验结果说明能适用于一般高效液相色谱。该气动活塞泵工作压力是在10~100公斤/匣米~2以内,流量在0~12毫升/分范围内可任意调节,且具有高压无脉动和流量较稳定的性能。
High performance liquid chromatography is a new technology of modern separation and analysis, which has been widely used in many fields. At present there are many analytical, preparative and microanalysis HPLC, but these instruments are more expensive. In this paper, a simple HPLC system consisting of a simple pneumatic piston pump, a stainless steel column with an inner diameter of 10 mm, silica gel H as a filler and a single wavelength (254 nm) UV detector is proposed. Experimental results show that it can be applied to general high performance liquid chromatography. The pneumatic piston pump working pressure is 10 ~ 100 kg / box meter ~ 2 within the flow rate of 0 to 12 ml / min range can be adjusted, and has no pulse of high pressure and flow more stable performance.
