目的:采用HPLC测定当归鲜品与干品中游离阿魏酸和总阿魏酸的含量,并对其进行比较。方法:ZORBAX EclipseXDB-C18(4.6 mm×250 mm,5μm)分析柱,流动相乙腈-0.085%磷酸(17∶83),检测波长316 nm,柱温35℃,流速1.0 mL·min-1,进样量为10μL。测定样品中游离阿魏酸和总阿魏酸的含量。结果:游离阿魏酸和总阿魏酸分别在0.024 2~0.121 0μg和0.0242~0.192 36μg呈良好的线性关系。回收率分别为96.34%,97.35%;09年当归干品中游离阿魏酸含量最高,鲜品中总阿魏酸含量最高。结论:该方法准确简便,具有相对良好的重复性和稳定性,有利于提高当归药材的质量控制。
Objective: To determine the content of free ferulic acid and total ferulic acid in fresh and dried Angelica sinensis Angelica samples by HPLC and to compare them. METHODS: ZORBAX EclipseXDB-C18 (4.6 mm × 250 mm, 5 μm) analytical column was used. The mobile phase consisted of acetonitrile and 0.085% phosphoric acid (17:83). The detection wavelength was set at 316 nm. The sample volume is 10 μL. Determination of the sample free ferulic acid and total ferulic acid content. Results: There was a good linear relationship between free ferulic acid and total ferulic acid between 0.024 2 and 0.121 0 μg and between 0.0242 and 0.192 36 μg. The recoveries were 96.34% and 97.35% respectively. In 2009, Angelica sinensis had the highest content of free ferulic acid and the highest content of ferulic acid in fresh products. Conclusion: The method is accurate and simple, with relatively good repeatability and stability, which is beneficial to improve the quality control of Angelica sinensis.
