目的研究细胞容积比值(cell volume ratio,CVR)对人结肠上皮NCM460细胞电阻抗谱的影响。方法采用Agilent 4294A阻抗分析仪测量了不同浓度NCM460细胞的电阻抗值和相位角,经过幅-频曲线和相-频曲线以及Nyquist图的综合分析,提取NCM460细胞的特征参数,观察细胞容积比值对NCM460细胞电生理性能的影响。结果 1)细胞容积比值对阻抗幅值最大增量(ΔZmax)、相位角峰值(θp)、低频阻抗值(Re ZL)和阻抗虚部峰值(Im Zp)等均有影响,并它们与细胞容积比值之间呈线性相关关系;2)特征频率(fc)与细胞容积比值无关,它来源于细胞膜的界面极化效应,是NCM460细胞对交流电场响应的电阻抗谱标志性参数。结论电阻抗谱方法能够获取NCM460细胞电学特征,为进一步的结肠癌细胞电生理研究提供了观测指标与方法。
Objective To study the effect of cell volume ratio (CVR) on the electronic impedance spectroscopy of human colonic epithelial cells (NCM460). Methods The electric impedance and phase angle of NCM460 cells were measured by Agilent 4294A impedance analyzer. The characteristic parameters of NCM460 cells were extracted by comprehensive analysis of amplitude-frequency curve, phase-frequency curve and Nyquist plot. Results 1) The ratio of cell volume affected the maximum increment of impedance (ΔZmax), phase angle (θp), low frequency impedance (Re ZL) and impedance peak (Im Zp) 2) The characteristic frequency (fc) is independent of cell volume ratio. It is derived from the interface polarization effect of cell membrane and is a landmark parameter of electrical impedance spectroscopy of NCM460 cells responding to AC electric field. Conclusion The electrical impedance spectroscopy method can obtain the electrical characteristics of NCM460 cells and provide the observation indexes and methods for the further study of the electrophysiology of colon cancer cells.
