产乙醇杆菌YUAN-3T是以糖蜜废水为底物分离出的具有良好自凝集现象的高效产氢菌株,在连续流反应器中减少了产氢细菌的流失,客观上起到了固定化的作用,保证了反应器内产氢细菌的高浓度,提高了糖蜜的利用率,降低了制氢成本,进一步加快了发酵法生物制氢技术的工业化进程.为了提高菌体产氢能力,并逐步阐明YUAN-3T在包括糖蜜在内的多种基质中发酵的活力影响因素,通过发酵罐静态试验及优化的氢化酶活性检测方法,分析了pH对YUAN-3T产氢能力及氢化酶活性相关性.结果表明,YUAN-3T的产氢能力对pH变化极为敏感,生物量在第9h 即pH开始快速下降时进入生长对数期,pH降为4时发酵缓慢,产气基本停止,生物量在24h达到最大值后开始减少.氢化酶活性随代谢过程迅速升高并在pH4.5时达到最大值后下降.
