目的:探讨JAK-STAT信号转导通路中转录因子GATA3在新疆维吾尔族寻常性银屑病患者皮损组织中的表达水平及其意义。方法:分别提取6例新疆维吾尔族寻常性银屑病患者皮损组织(银屑病组)和5例新疆维吾尔族正常人皮肤组织(正常对照组)的总RNA,反转录合成cDNA,用RT2 profiler PCR Array检测JAK/STAT信号通路相关基因的mRNA表达水平,通过在线软件对数据进行分析,并采用免疫组化方法检测45例银屑病组和21例正常对照组中GATA3蛋白水平的表达情况,并进行相关分析。结果:新疆维吾尔族银屑病患者皮损组织与正常人皮肤组织对比,共有17个表达差异2倍以上且P<0.05的基因,其中GATA3 mRNA在银屑病组皮损组织中的表达水平比正常对照组表达水平下降。GATA3蛋白在正常对照组中为表皮全层细胞核表达,在银屑病组皮损组织中表达量均较正常对照组明显降低(P<0.001)。结论:GATA3在新疆维吾尔族寻常性银屑病患者皮损表皮中表达明显下调,提示GATA3可能在新疆维吾尔族银屑病发病机制中发挥作用。“,”To investigate the expression and significance of the transcription factor GATA3 of JAK-STAT sig-naling pathway in lesions of psoriasis vulgaris Uygur patients in Xinjiang. Methods: The total RNA were extracted from 6 patients with psoriasis vulgaris and 5 healthy subjects and reversely transcribed in cDNA. RT2 profiler PCR array was used to detect the mRNA expression level of JAK/STAT signaling pathway related genes , and the expression of GATA3 proteins were detected by immunohistochemistry in 45 patients with psoriasis vulgaris and 21 healthy controls. Results: Compared with con-trol group, 17 candidate genes were identified that differentially expressed in psoriasis patients. The expression level of GATA3 mRNA existing in Xinjiang Uygur psoriatic lesions was down-regulated compared with the control group. The GATA3 protein were expressed at full-thickness epidermal cell nucleus in normal controls, while in tissue lesions of the pilot group the ex-pression were evidently decreased. There was statistical significance between those two groups (P<0.001). Conclusion: The ex-pression of GATA3 is significantly down-regulated within psoriasis epidermis in Xinjiang Uygur, which suggests that GATA3 may play a part in the pathogenesis of the Uygur psoriatic.
