目的:研究掌叶蝎子草Girardinia diversifolia(Link)Friis多糖对糖尿病小鼠的降糖效果及体内外抗氧化活性.方法:在体外化学模拟条件下,测定掌叶蝎子草粗多糖对超氧阴离子(O2-·)、羟基自由基(.OH)的清除活性;采用四氧嘧啶诱导建立糖尿病小鼠模型,糖尿病小鼠分别灌胃盐酸二甲双胍、掌叶蝎子草多糖低剂量(200mg/kg)、高剂量(400mg/kg),给药后测定小鼠的空腹血糖值,血清MDA体积分数和SOD的活性.结果:掌叶蝎子草粗多糖清除·OH的IC50值≥2mg/mL,清除O2-·的IC50值≥5mg/mL,掌叶蝎子草粗多糖清除自由基活性与其体积质量分数成正比.掌叶蝎子草高剂量多糖能降低糖尿病小鼠的血糖值、采食量和饮水量,提高了小鼠胸腺指数和脾脏指数,升高血清SOD水平,降低血清MDA水平,抑制糖尿病小鼠体质量减轻,但治疗效果略低于阳性药物盐酸二甲双胍片;掌叶蝎子草多糖低剂量降血糖效果不明显.结论:掌叶蝎子草多糖具有降血糖作用,这为蝎子草多糖有可能成为新的天然降糖药物或保健品提供了科学依据.
Objective: To study the hypoglycemic effect and anti-oxidative activity of polysaccharide from Girardinia diversifolia (Link) Friis polysaccharide on diabetic mice in vitro.METHODS: The crude polysaccharide of Scirpus molitor Oxygen free radical (OH) scavenging activity was established. Alloxan-induced diabetes mice model were established. The diabetic mice were treated with metformin hydrochloride, low dose (200mg / kg) (400mg / kg), the fasting blood glucose, the content of serum MDA and the activity of SOD in the mice were determined after administration.Results: IC50 value of crude polysaccharide scavenging · OH of Scirpus sonata ≥2mg / mL, clearing O2- · The IC50 value of ≥5mg / mL, the free radical scavenging activity of crude polysaccharide Scorpion grass is proportional to the volume fraction of Scorpion grass high-dose polysaccharide can reduce blood glucose in diabetic mice, feed intake and drinking water, increased Mouse thymus index and spleen index, increased serum SOD level, reduced serum MDA level, inhibited the weight loss of diabetic mice, but the therapeutic effect was slightly lower than the positive drug metformin hydrochloride tablets; low dose of Scirpus grass polysaccharides hypoglycemic effect is not obvious. Conclusion: The polysaccharide from Scirpus monsiera has hypoglycemic effect, which provides a scientific basis for the scorpion polysaccharides to become a new natural drug or health supplement.
