目的:优选大孔吸附树脂分离、纯化甘草总黄酮的工艺条件。方法:以总黄酮吸附量和解吸率为考察指标,对12种不同类型树脂进行筛选,在此基础上对其纯化工艺条件进行优选。结果:AB-8型大孔吸附树脂对甘草总黄酮的纯化效果最好,其最佳工艺条件为上样液质量浓度为0.2 g.mL-1,每mL树脂最大上样量为2 mL,吸附流速0.5 BV.h-1,上样液pH 6,6BV水以流速1 BV.h-1洗脱除杂,4 BV 70%乙醇以1 BV.h-1流速洗脱。该优选条件下,洗脱物中甘草总黄酮纯度为38%。结论:该优选工艺合理、可行,适合工业生产。
Objective: To optimize the conditions for the separation and purification of total flavonoids from licorice by macroporous adsorption resin. Methods: Based on the total amount of flavonoids adsorbed and desorption rate, twelve different types of resins were screened. Based on these results, the purification process conditions were optimized. Results: The results showed that AB-8 macroporous resin had the best purification effect on total flavonoids of Glycyrrhiza uralensis Fisch. The optimum conditions were as follows: the mass concentration of sample solution was 0.2 g.mL-1, the maximum sample volume per mL was 2 mL, Adsorption flow rate of 0.5 BV.h-1, the sample solution pH of 6, 6 BV water at a flow rate of 1 BV.h-1 elution impurity removal, 4 BV 70% ethanol at 1 BV.h-1 flow rate elution. Under the preferred conditions, the licorice flavonoids purity of the eluate is 38%. Conclusion: The optimal process is reasonable and feasible, suitable for industrial production.
