目的建立一种应用超高效液相色谱法同时测定饮料中14种着色剂、甜味剂和防腐剂的分析方法。方法样品经ZOXBAX SB-C_(18)色谱柱分离,以甲醇和100mmol/L磷酸二氢铵溶液为流动相进行梯度洗脱,流速0.2mL/min,柱温35℃。采用二极管阵列检测器,在190~600nm波长下进行扫描,230nm和200nm波长下进行检测,外标法定量。结果在最佳色谱条件下,这14种食品添加剂可在20min内有效分离,标准曲线线性关系良好(r>0.996)。平均回收率为92.7%~97.1%,相对标准偏差为0.61%~2.54%。当取样量为5g,稀释体积为25mL时,检出限(limits of detection,LODs)为0.3~0.6mg/kg。结论建立的方法可实现对饮料中多种着色剂、甜味剂和防腐剂的同时检测,具有快速、准确、灵敏的特点。
Objective To establish a method for the simultaneous determination of 14 colorants, sweeteners and preservatives in beverages by ultra performance liquid chromatography. Methods The samples were separated on a ZOXBAX SB-C 18 column and eluted with methanol and 100 mmol / L ammonium dihydrogen phosphate solution as mobile phase with a flow rate of 0.2 mL / min and a column temperature of 35 ℃. Using diode array detector, scanning at 190 ~ 600nm wavelength, 230nm and 200nm wavelength detection, external standard method. Results Under the optimal chromatographic conditions, the 14 kinds of food additives can be effectively separated in 20 minutes, and the standard curve has a good linear relationship (r> 0.996). The average recoveries ranged from 92.7% to 97.1% with relative standard deviations of 0.61% to 2.54%. The limits of detection (LODs) ranged from 0.3 to 0.6 mg / kg when the sample volume was 5 g and the dilution volume was 25 mL. Conclusion The established method can detect many kinds of colorants, sweeteners and preservatives in the beverage at the same time. It is fast, accurate and sensitive.
