目的 探讨镉 (Cd)致淋巴细胞DNA损伤及其镉免疫毒性机制中的地位和作用。方法 运用单细胞凝胶电泳法、MTT法和荧光抗体标记法分别检测各染镉剂量组的DNA损伤、T淋巴细胞增殖转化功能和T淋巴细胞亚群情况。结果 各染镉组的DNA损伤与对照组有显著性差异 (P <0 .0 5 ) ;T淋巴细胞增殖功能与对照比较有降低 (P <0 0 5 ) ,并均随镉剂量的升高有逐步加重的趋势 ;DNA损伤与T淋巴细胞增殖转化功能有显著的相关关系 (Pearson相关分析 ,r =0 .80 78,P <0 .0 1)。结论 镉的免疫毒性机制中镉致淋巴细胞DNA损伤可能是重要的机制之一。
Objective To investigate the role and status of cadmium (Cd) -induced lymphocyte DNA damage and its cadmium immunotoxicity mechanism. Methods Single cell gel electrophoresis, MTT assay and fluorescent antibody labeling method were used to detect DNA damage, T lymphocyte proliferation and transformation function and T lymphocyte subsets in each cadmium dose group. Results DNA damage in each cadmium group was significantly different from that in the control group (P <0.05). The proliferation of T lymphocytes was significantly lower than that of the control group (P <0.05) (P <0.01). There was a significant correlation between DNA damage and the proliferation and transformation of T lymphocytes (Pearson correlation analysis, r = 0.8078, P <0.01). Conclusion Cadmium-induced lymphocyte DNA damage may be one of the important mechanisms in the mechanism of cadmium immunotoxicity.
