目的 研究人工合成的高活性心钠素 (hANP)对妊高征患者一氧化氮 (NO)及内皮素 (ET)水平的影响。方法 对 45例妊高征患者随机分为hANP组 30例及安慰剂组 15例 ,分别给予每公斤体重hANP 6 μg及安慰剂静滴 30min。应用活化镉还原剂与Griess反应结合法及放射免疫分析法分别测定用药前后血清NO代谢终产物硝酸基 (NO3-)及血浆ET水平 ,并观察血压变化。结果 hANP使NO3-水平升高 ,有显著差异 (P <0 0 1) ,ET呈下降趋势 ,但无显著差异 (P >0 0 5 ) ,NO3-/ET比值升高 ,平均动脉压 (MAP)下降均有显著差异 (分别 P <0 0 1) ,MAP下降值与NO3-及ET变化值均无显著相关性 (P >0 0 5 )。安慰剂组MAP、NO3-及ET均无明显变化。结论 hANP影响妊高征患者NO合成及ET分泌水平 ,可能在改善妊高征病理生理变化中具有重要作用
Objective To investigate the effects of synthetic high activity atrial natriuretic peptide (hANP) on the levels of nitric oxide (NO) and endothelin (ET) in patients with pregnancy induced hypertension. Methods Forty-five PIH patients were randomly divided into hANP group (n = 30) and placebo group (n = 15). HANP 6 μg / kg body weight and placebo infusion for 30 min were given respectively. The activity of nitric oxide (NO3-) and plasma ET in the end products of serum NO metabolites before and after treatment were determined by using activated cadmium reductant and Griess reaction combined with radioimmunoassay. The changes of blood pressure were observed. Results There was a significant difference (P <0.01) between hANP and NO levels, but there was no significant difference between the two groups (P> 0.05), the NO3- / ET ratio increased, mean arterial pressure ) (P <0.01, respectively). There was no significant correlation between the decrease of MAP and the change of NO3- and ET (P> 0.05). There was no significant change in MAP, NO3- and ET in the placebo group. Conclusions The effect of hANP on NO synthesis and ET secretion in patients with PIH may play an important role in the pathophysiological changes of PIH
