目的探讨天然化合物长梗秦艽酮对吉西他滨耐药肺腺癌细胞H1975增殖、凋亡的影响及其可能机制。方法采用MTT比色法检测长梗秦艽酮对H1975细胞增殖的影响;采用AnnexinV/PI流式细胞仪双染法检测长梗秦艽酮诱导H1975细胞凋亡;通过荧光电子显微镜观察长梗秦艽酮诱导凋亡的细胞核形态变化;通过Western blot研究长梗秦艽酮发挥抑癌作用的靶蛋白及机制。结果长梗秦艽酮显著抑制肺癌细胞H1975的增殖,且呈现明显时间剂量依赖性;长梗秦艽酮可以诱导细胞凋亡;长梗秦艽酮处理使细胞出现染色质边集、核碎裂等典型的细胞凋亡改变;长梗秦艽酮处理使Bcl-2表达降低,Bax表达升高。结论长梗秦艽酮可以抑制H1975细胞增殖,通过对Bcl-2和Bax的调节诱导细胞凋亡,是一种潜在的天然抗肿瘤药物。
Objective To investigate the effects of long-acting genistein on the proliferation and apoptosis of human lung cancer cell line H1975 and its possible mechanism. Methods MTT colorimetric assay was used to detect the effect of long-stemmed genistein on the proliferation of H1975 cells. The apoptosis of H1975 cells was detected by Annexin V / PI dual-flow cytometry. The induction of long-stemmed genistein by fluorescence electron microscopy Apoptotic nuclear morphological changes; Western blot study of long-term Qin Qin ketone play a role in tumor suppressor target protein and mechanism. Results Long Qin Qin ketone can significantly inhibit the proliferation of lung cancer cells H1975, and showed a significant dose-dependent manner; long-Qin Qin ketone can induce apoptosis; long-stemmed Qin ketone ketone cells appear chromatin edge set, nuclear fragmentation and other typical Apoptosis changes; Long Qin Qin ketone treatment decreased Bcl-2 expression, Bax expression increased. Conclusion Long Qin Qin ketone can inhibit the proliferation of H1975 cells, which is a potential natural anticancer drug through the regulation of Bcl-2 and Bax.
